Hexokinase is involved in glycolysis. It catalyses the first step by transferring a phosphate from ATP to its substrate glucose, forming glucose-6-phosphate. Thus, the amount of converted substrate is directly related to the amount of ADP produced. You want to investigate the kinetics of hexokinase that follows the Michaelis-Menten scheme. In order to determine the maximal reaction velocity (Vmax) and the Michaelis-Menten constant (KM), you need to measure the reaction velocity (V0) at different substrate concentrations ([S]).

You are given two csv data files:
• ADP_fluorescence_kits.csv – Data from the preliminary experiments of measuring the fluorescence emission in dependence of the ADP concentration (µM).
• enzyme_kinetics.csv – Data obtained with the kit to determine the reaction velocities (U/mg) for different substrate concentrations (µM).
Analysis Task
As a preliminary test you want to check if the fluorescence emission has a linear correlation to the ADP concentration. You make up solutions with different ADP concentrations and measure the fluorescence emission for each of the kits. The data is stored in the ADP_fluorescence_kits.csv data file.
The following questions are a guide to your analysis. Remember that your report should be written as described in the STAT ICA2 Guidelines and you should not provide numbered answers to these tasks in your report.
1. Load the ADP_fluorescence_kits.csv file into a data frame named ADP_kits. Describe how many kits have been assessed, the different emission wavelengths, the different ADP concentrations (µM) used and number of replicates.
2. Examine the data by providing a graphical representation of the association between fluorescence emission and the ADP concentration and the correlation coefficient for each kit. Which kit shows an approximately linear relationship between these two variables?
3. Identify which kit is suitable to determine the concentration of ADP in an enzymatic assay.
4. Use the data of the most suitable kit to plot a standard curve. What is the dependent and what is the independent variable?
5. Perform linear regression on the data and store the output in a variable named reg. Report the linear equation of the line fitted.
6. Plot the points of the standard curve and the fitted line in blue to visually assess how good the fit is. Provide also a quantitative measure of the goodness of the fit.
